Kujo Jotaro, sıradan ve popüler bir Japon lise öğrencisidir; ta ki bir ruh tarafından ele geçirildiğini düşünüp kendini hapse atana kadar. Büyükbabası Joseph Joestar'ı görüp arkadaşı Muhammed Abdul ile dövüştükten sonra Jotaro, "Ruh"un aslında Star Platinum, yani yarı katı bir form almış dövüş enerjisi olduğunu öğrenir. Daha sonra annesi bir Direnç kazanır ve hastalanır. Jotaro, bunun sebebinin vampir Dio Brando'nun, Jotaro'nun büyük büyükbabası Jonathan Joestar'a yenilmesinden 100 yıl sonra yeniden canlanması olduğunu öğrenir. Jotaro, Dio'yu kesin olarak yenmek için Joseph ve Abdul ile birlikte Mısır'a gitmeye karar verir.
